Currently, there are many tissue optical clearing methods available worldwide. Some of the techniques are favorable for preserving the fluorescence labels, while some of them maximize the transparency of the tissue. However, there is no optical clearing method that is proved to be optimized for photoacoustic imaging, which is a novel label-free high-resolution imaging technology. To this end, this research project aims to find out the optimal one from the existing list. The hypothesis will be tested with our photoacoustic microscope.
Students are required to take safety training courses, including Chemical Safety I and II, Laser Safety, and Biological Safety.
The applicant should be able to analyze different optical clearing methods and choose the most appropriate one for photoacoustic microscopy. After the search, the applicant should try out the chosen clearing method and validate the hypothesis with the existing photoacoustic microscope in my laboratory.
By doing background literature search with the ultimate goal of integrating optical clearing and photoacoustic microscopy, the applicant will be able to understand the research fields in both areas and also strengthen his/her critical thinking ability. The applicant will also have a chance for performing hands-on experiments, trying different optical clearing methods with our photoacoustic microscope.
